After completing his postdoctoral training, Dr. Shi became an Instructor (1997) and then Assistant Professor (1999) at the Baylor College of Medicine in Houston, Texas. His research focused on the molecular biology of glutathione (GSH), a dominant intracellular antioxidant. Dr. Shi and his colleagues first identified mutations in GSH synthetase as the cause of the disease 5–oxoprolinuria, a rare but serious human genetic disorder. Using knockout technology, they first proved that GSH is essential for mammalian development by dispensing it in cultured mammalian cells. In 2000, Dr. Shi joined Lexicon Genetics (The Woodlands, TX) where he established and oversaw a large-scale phenotypic analysis of knockout mice. As a Senior Scientific Group Leader, he also developed the use of small animal imaging systems including magnetic resonance imaging (MRI) and micro–computed tomography (micro–CT) for phenotypic analysis and pre-clinical testing. He joined the Methodist Hospital Research Institute in 2007 as Chief of Small Animal Imaging and a Research Scientist in the Department of Radiology.
The long-term goals of Dr. Shi´s research are: (i) to discover novel cellular proteins/pathways that can serve as therapeutic targets for human diseases including cancer, or as targets for molecular imaging; and (ii) to support the development of novel molecular imaging agents/methods by providing expertise and core services in molecular biology and small animal imaging. His laboratory has recently discovered a novel Obg–like ATPase (OLA1) which functions as a regulator of oxidative stress response and many other cellular functions through translational and post-translational pathways. His group is currently focusing on demonstration of in vivo physiological functions of OLA1 and validation of OLA1 as an anti-cancer or anti-metastasis target.
